Costa-Lotufo, et al. 4: Journal: Oxidative Medicine and Cellular Longevity 5: Date: 2020 6: ## Abstract 7: Amikacin (AMA) is an aminoglycoside antibiotic that results in nephrotoxicity that affects mainly renal proximal tubule epithelial cells (RPTEC); however, there are few reports explaining the mechanism of AMA-induced cytotoxicity on RPTEC. The aim of this study was to evaluate lycopene against AMA-induced cytotoxicity via modulation of reactive oxygen species (ROS) production and mitochondrial membrane potential (MMP) in RPTEC cells by using a cell viability assay (MTT), intracellular ROS content measured by H2DCF-DA fluorescent probe, MMP measured by JC-1 dye, the expression of p53-related gene (Bax), and caspase-3 assay. Increased AMA concentrations induce serious mitochondrial dysfunction; therefore, inhibiting ROS production might be associated with protecting cell viability by preserving MMP; as a result, preserving MMP preserves cell viability against AMA-induced toxicity. The results showed that the application of concentrations taken up by cells from lycopene enriched milk (LEM) (10 μg/ml) ameliorated AMA (640 μM)-induced cell viability loss; however, high concentration (100 μg/ml) reversed cell viability similar to the control cells. Moreover, lycopene-treated cells showed high mitochondrial membrane status compared to AMA-treated cells. Furthermore, lycopene ameliorated AMA-induced increase in both intracellular ROS production and Bax level but enhanced expression of caspase-3 level. These results represent that lycopene has anti-toxic effects against AMA-induced cytotoxicity through ROS inhibition and caspase-3 activation effects. 8: ## 1. Introduction 9: Aminoglycoside antibiotics are an extensively used group of antibiotics used for severe Gram-negative infections [[1], [2]]. Amikacin (AMA) is an aminoglycoside antibiotic that has a broader spectrum for treatment of bacterial infection than other aminoglycoside antibiotics [[1]]. Additionally, it can pass through human placental barrier without affecting fetus [[3]]. Amikacin is considered as one of the major causes of renal damage associated with aminoglycosides antibiotic use [[4]]. Although several molecular mechanisms proposed for aminoglycoside-induced nephrotoxicity in human cases [[5]] and experimental studies [[6]– [9]], detailed mechanisms have not clearly been discovered yet. 10: Oxidative stress is thought to play an essential role in many types of renal injury induced by diverse toxins including anticancer drugs [[10], [11]] as well as aminoglycoside antibiotics [[12], [13]].
